NMN Side Effects: What the Research Actually Says (UK Guide 2026)

If you've searched "NMN side effects" you probably want one of two things: reassurance that you're not about to do something harmful, or a clear-eyed read on what could go wrong before you spend money. This guide gives you both, based on what the published human clinical trials actually report.

We'll be upfront: Ivvion makes NMN. We have a commercial interest in you not being scared off the category. So we've written this to be useful even if you ultimately decide NMN isn't for you, or decide to buy from a competitor.

The short answer

In the published human clinical trials, NMN has been very well tolerated. Across multiple peer-reviewed studies at doses up to 1,250 mg per day, no serious adverse events have been attributed to NMN supplementation. Reported side effects have been mild and similar to placebo — occasional gastrointestinal discomfort, mild flushing, headache.

NMN is not known to cause major drug interactions, but it has not been formally tested with every prescription medication. If you take regular medication, particularly for blood pressure, blood thinning, diabetes, or any serious condition, you should talk to your GP before starting.

The honest caveat is that long-term safety data (multi-year human supplementation) is still limited. NMN has only been a mass-market supplement for about six years. Studies are ongoing.

What side effects were reported in the human trials?

Yoshino et al, 2021, Science — 250 mg/day for 10 weeks, postmenopausal women. No serious adverse events. Reported mild side effects were similar between the NMN group and placebo group. The trial monitored liver function, kidney function, lipid panels, and complete blood counts — nothing showed clinically meaningful change in the NMN group.

Igarashi et al, 2022, npj Aging — 250 mg/day for 12 weeks, older adults. No serious adverse events. Mild gastrointestinal complaints were the most commonly reported, at similar rates in NMN and placebo groups.

Liao et al, 2023, Journal of the International Society of Sports Nutrition — 300 mg/day for 6 weeks, recreational runners. No serious adverse events. No clinically meaningful changes in standard blood biomarkers.

Various safety studies, 2019–2024. Single-dose pharmacokinetic studies at doses up to 1,250 mg in healthy adults. Mild headache and flushing were the most commonly reported short-term effects. No serious events.

The pattern across all of these is the same: NMN at the doses tested is a very well-tolerated molecule.

What about the worried-internet side effects?

"NMN causes cancer"

This is the loudest concern online, and it deserves a careful answer.

The concern stems from the fact that NAD+ is used by many cells, including cancer cells, to maintain energy and DNA repair. The theoretical worry is that supplementing NMN could provide cancer cells with more of the resource they need to grow.

In laboratory and animal models, the picture is mixed and depends heavily on the type of cancer cell and the experimental setup. In humans, no clinical trial has reported NMN increasing cancer risk. But no clinical trial has been designed specifically to detect cancer signals over decades of supplementation either.

What this means practically:

  • If you have an active cancer diagnosis, do not take NMN without discussing it with your oncologist.
  • If you have a family history of cancer or you're at high risk, talk to a GP before starting.
  • If you're a healthy adult with no cancer concerns, the current evidence does not support avoiding NMN on cancer grounds — but be aware the long-term safety data is incomplete.

"NMN raises homocysteine"

Some NAD+ precursors theoretically affect the methylation cycle, which can affect homocysteine levels. In the published human trials, no clinically meaningful homocysteine change has been attributed to NMN. If you're concerned, ask your GP for a homocysteine blood test before starting and again at 12 weeks.

"NMN causes insomnia"

Anecdotally, a minority of users report difficulty falling asleep if they take NMN late in the day. The practical solution: take NMN in the morning. The clinical trials all used morning dosing.

"NMN causes anxiety or restlessness"

Again, anecdotal in a minority of users. If you notice this, lower the dose or stop. There's no reported pattern of NMN causing clinically significant anxiety in the trials.

"NMN is a hidden steroid or hormone"

It isn't. NMN is a nucleotide, not a hormone. There's no steroidal or hormonal pathway involved.

Who should not take NMN?

Pregnant or breastfeeding women. No clinical trials have tested NMN in pregnancy or lactation. Default to not taking it.

Children and teenagers under 18. Same reason — no safety data in this group.

People with active cancer. Talk to your oncologist.

People on chemotherapy, immunosuppressants, or other powerful medications. Talk to your prescribing doctor.

People who experience a clear adverse reaction. Stop and ask a GP.

People who feel uneasy about taking it. Don't take it. The case for NMN is modest measurable benefits in healthy adults over 40. It's not a treatment for anything.

Drug interactions to be aware of

NMN has no documented major drug interactions, but here are the medication categories where it's worth a quick GP conversation:

  • Blood pressure medications — a check-in is sensible
  • Diabetes or metformin — NMN improved insulin sensitivity in the Yoshino trial; your dose may need adjusting
  • Anticoagulants (warfarin, DOACs) — standard supplement caution applies
  • Statins — no documented interaction
  • SSRIs or antidepressants — no documented interaction
  • Immunosuppressants — talk to your prescribing doctor

Quality issues that matter

Most bad experiences with NMN aren't side effects of NMN — they're side effects of bad NMN supplements.

Contamination. Low-quality NMN from untested supply chains can contain trace contaminants. The protection is third-party testing — buy from brands that publish their test certificates.

Purity below 98%. NMN that's actually 60% NMN and 40% nicotinamide produces different effects than pure NMN.

Wrong dose. A bottle that claims 500 mg per serving but delivers less due to filler dilution produces no effect — which buyers then attribute to "NMN doesn't work."

Degradation. NMN is moisture-sensitive. Store in a cool, dry place and finish a bottle within 90 days of opening.

How to minimise your personal risk

  1. Start at the lower end of the trial dose range (250 mg/day) and stay there for 8 weeks before changing anything.
  2. Buy from a brand that publishes a third-party test certificate and lists the named raw material (e.g. Uthever®).
  3. Take it in the morning, not before bed.
  4. Have a baseline GP check-in if you take medication or have a condition.
  5. Listen to your body. If something feels off in the first two weeks, stop.

Frequently asked questions

Is NMN safe to take long-term? No multi-year human safety data exists yet. Short-term trials (up to 12 months) show NMN is well-tolerated. Long-term studies are ongoing.

Can I take NMN every day? The trials used continuous daily dosing for weeks to months without adverse effects.

Are some NMN brands safer than others? The molecule is the same; the difference is in purity and contamination control. Third-party tested NMN from a GMP-certified manufacturer is meaningfully safer than untested bulk product.

Can NMN cause weight gain or weight loss? No clinically meaningful effect on body weight has been reported in the human trials.

Does NMN affect fertility? No human trial has been designed to test fertility outcomes. Default position: don't take NMN if you're trying to conceive, pregnant, or breastfeeding.

Should I take a break from NMN periodically? There's no evidence cycling is necessary.
